With a Galaxy Note 2, when answering a call with headphones (but not a headset) connected, the audio output goes through the headphones and the inbuilt microphone works as though it were a normal call (so it's too quiet when it's a few inches away). When I switch to speakerphone, it ignores the headphones entirely and audio output switches to inbuilt speaker.

Basically, I'd like to talk to people with my phone on my desk and a pair of conventional headphones.

You might want to take a look at SoundAbout. One of its promises:

DETECT or IGNORE that a headset has a microphone when plugged in. Only used if the headphone/headset plug in/out detection is set to ALLOW.

Which to me sounds like what you are looking for.

Unfortunately not. I bought the app, emailed the developer and it can't do it. He said it was a limitation of Android that you can't configure which microphone to use. – Ben Heley Nov 21 '12 at 19:04
That I would call "misleading advertisement", as the description clearly claims the app capable of this. So I'd ask for refund (at least), and to correct that claim. – Izzy Nov 21 '12 at 20:13
